A Diarylheptanoid Exhibits Protective effects against Amyloid-β Induced Cytotoxicity

چکیده
Alzheimer’s disease (AD) is a severe neurodegenerative disorder characterized by β-amyloid (Aβ) plaques, neurofibrillary tangles, neuronal death, progressive cognitive impairment and memory loss. β-amyloid peptides (Aβ) are major compositions of so-called senile plaques, which are hallmarks of AD and contribute to AD progression. Therefore, it is important to discover therapeutics against Aβ-induced toxicity. Here, we report that 7(4Hydroxyphenyl)1phenyl-4E-hepten-3-one (AO-1), a diarylheptanoid extracted from the rhizomes of Alpinia officinarum, showed effects against Aβ-induced cytotoxicity. The results demonstrated that AO-1 possesses neuroprotective activities which are capable of maintaining normal cell viability of PC12 cells and primary cortical neurons after Aβ insult.

Screening seven Iranian medicinal plants for protective effects against β-Amyloid-induced cytotoxicity in cultured cerebellar granule neurons
Background and objectives: Alzheimer's disease (AD) as a neurodegenerative disorder is the most common form of dementia in the elderly. According to the amyloid hypothesis, accumulation of amyloid beta (Aβ) plaques, which are mostly constituted of Aβ peptide aggregates, triggers pathological cascades that lead to neuronal cell death. Thus, modulation of Aβ toxicity is the hopef...

Alzheimer’s disease (AD) is a neurodegenerative disease that was characterized with deposit of beta amyloid (Aβ) aggregate in senile plaque. Oxidative damage to neurons and loss of cholinergic neurons in forebrain region are observed in this disease. Melissa officinalis is a medicinal plant from Lamiaceae family, used traditionally in the treatment of cognitive disorders. It has cholinomimeti...

Background: Our previous findings indicated that carvacrol and thymol alleviate cognitive impairments caused by Aβ in rodent models of Alzheimer's disease (AD). In this study, the neuroprotective effects of carvacrol and thymol against Aβ25-35-induced cytotoxicity were evaluated, and the potential mechanisms were determined. Methods: PC12 cells were pretreated with Aβ25-35 for 2 h, followed by ...

Objective(s): In previous studies, researchers observed that doxepin could improve cognitive processes and has protective effectson the central nervous system. Thus, this study was designed to analyze the effects of doxepin on β-amyloid (Aβ)-induced memory impairment and neuronal toxicity in ratand to explore the underlying mechanism. Materials and Methods: Rats were treated with Aβ1-42 and dox...
